Detection of IL‑10 in Human Blood Lymphocytes by Flow Cytometry. Human peripheral blood lymphocytes untreated (A) or treated (B) with PMA and Calcium Ionomycin were stained with Mouse Anti-Human IL‑10 APC‑conjugated Monoclonal Antibody (Catalog # IC2172A) and Mouse Anti-Human CD3 epsilon PE‑conjugated Monoclonal Antibody (Catalog # FAB100P). Quadrant markers were set based on control antibody staining (Catalog # IC002A). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Interleukin-10 (IL-10) is a pleiotropic cytokine that can exert either immunostimulatory or immunosuppressive effects on a variety of cell types. It is produced predominantly by Th2 lymphocytes and other activated lymphoid cells.
