K562 human cell line was stained with Rat Anti-Human IRAK Monoclonal Antibody (Catalog # MAB39191, filled histogram) or isotype control antibody (Catalog # MAB005, open histogram) followed by anti-Rat IgG APC-conjugated secondary antibody (Catalog # F0113). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). IRAK4 was detected in immersion fixed U937 human histiocytic lymphoma cell line using Rat Anti-Human IRAK4 Monoclonal Antibody (Catalog # MAB39191) at 8 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rat IgG Secondary Antibody (red; Catalog # NL013) and counterstained with DAPI (blue).
