PDZK1 (PDZ Domain-containing 1; also CFTR-associated protein of 70 kDa/CAP70) is a 65 - 70 kDa member of the NHER (Na/H Exchange Regulator) family of proteins. It is only expressed in epithelium, particularly that found in intestine, kidney and liver. PDZK1 serves as a scaffold protein that connects membrane proteins to cytoplasmic regulators. Human PDZK1 is 519 amino acids (aa) in length. It contains four PDZ domains (aa 9 - 90; 134 - 215; 243 - 323; and 378 - 458) plus two phosphorylation sites at Ser492 and Ser514. There are two potential truncated forms of human PDZK1, one that ends after Leu330, and a second that ends after Lys264. Human PDZK1 shares 78% aa identity with mouse PDZK1.

Detection of Human PDZK1 by Western Blot.
Western blot shows lysates of Calu-6 human lung carcinoma cell line, U266 human myeloma cell line, MCF-7 human breast cancer cell line, and mouse liver tissue. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse PDZK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4997) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for PDZK1 at approximately 80 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.PDZK1 in Human Kidney.
PDZK1 was detected in immersion fixed paraffin-embedded sections of human kidney using Sheep Anti-Human/Mouse PDZK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4997) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Specific labeling was localized to the brush border of proximal tubules.
