Detection of Human SOCS‑6 by Western Blot. Western blot shows lysates of Raji human Burkitt's lymphoma cell line, Daudi human Burkitt's lymphoma cell line, Jurkat human acute T cell leukemia cell line, NC-37 human Burkitt's lymphoma B lymphoblast cell line, and MOLT-4 human acute lymphoblastic leukemia cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human SOCS-6 Monoclonal Antibody (Catalog # MAB5808) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for SOCS-6 at approximately 75 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

SOCS‑6 in Jurkat Human Cell Line. SOCS-6 was detected in immersion fixed Jurkat human acute T cell leukemia cell line using Mouse Anti-Human SOCS-6 Monoclonal Antibody (Catalog # MAB5808) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cell surfaces. View our protocol for Fluorescent ICC Staining of Non-adherent Cells.

Background: SOCS-6
SOCS-6 (suppressor of cytokine signaling 6; also CIS-4) is an ~60 kDa member of the SOCS family of proteins. SOCS-6 can be found in either the cytoplasm or nucleus of hematopoietic progenitors. The 535 amino acid (aa) human SOCS-6 contains one SH2 domain (aa 384‑491) and a SOCS box (aa 486‑535), which recruits E3 ubiquitin ligase complexes. It is reported to antagonize IRS-4, PI3K p85, STAT3, and other SOCS proteins, probably by promoting their degradation. SOCS-6 shares 84% and 82% aa identity with mouse and rat SOCS-6, respectively. An isoform of 418 aa diverges after aa 404.
