Background: TGF-beta RI/ALK-5

Human TGF-beta RI (also ALK5) is a 55 kDa member of the Ser/Thr kinase family of transmembrane proteins. It is a type I membrane glycoprotein that is 479 amino acids (aa) in length. The short 101 aa extracellular domain (ECD) contains multiple cysteines and one potential N-linked glycosylation site. It is ubiquitously expressed, and forms a functional heterotetrameric TGF-beta receptor with TGF-beta RII. TGF-beta binding to TGF-beta RII phosphorylates TGF-beta RI, which then activates SMAD transcriptional proteins. The human TGF-beta RI ECD is 91%, 90%, 91%, and 93% aa identical to the TGF-beta RI ECD in pig, mouse, cow and rat, respectively.

    Immobilized TBRII onto ELISA plate. Then incubated plate with TGFb1 + TBRI solution. Detect binding of TBRI to TGFb1+TBRII complex by using Anti-TBRI-biotin. No binding of TBRI only to plate.
