Product Specifications

Immunogen

Recombinant full-length human p53 protein (Uniprot: P04637)

Localization

Predominantly nuclear

Specificity

Recognizes a 53kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53 protein. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.

Knockout Validated: p53 Antibody (TRP/817) [NBP2-44982]

Western Blot: p53 Antibody (TRP/817) [NBP2-44982]

Western Blot: p53 Antibody (TRP/817) [NBP2-44982] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and p53 knockout (KO) HeLa cell line. PVDF membrane was probed with 0.5 ug/ml of Mouse Anti-Human HSP27 Monoclonal Antibody (Catalog # NBP2-44982) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (catalog number HAF018). Specific band was detected for p53 at approximately 53 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

Applications for p53 Antibody (TRP/817)

Application
Recommended Usage

Immunohistochemistry-Paraffin

1-2 ug/ml

Western Blot

1-2 ug/ml
Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: p53

p53 is a stress-regulated transcription factor that was first identified as an SV40 large T antigen-binding protein. It plays a major role in the cellular response to DNA damage and other genomic aberrations. The activation of p53 can lead to either cell cycle arrest and DNA repair, or apoptosis. p53 is phosphorylated at multiple sites in vivo and by several different protein kinases in vitro. Mutation of p53 is the most common genetic change so far identified in a number of major carcinomas. This antibody can be used for the specific detection of human p53 that is synthesized in the presence of p53 from other species.
