Flow Cytometry: AHR Antibody [Biotin] [NB100-2289B] - An intracellular stain was performed on U-87 cells with AHR Antibody NB100-2289B (blue) and a matched isotype control (orange). Both antibodies were conjugated to Biotin. Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Streptavidin - R-Phycoerythrin Protein (2012-1000, Novus Biologicals).

Background: AHR

The Ah Receptor belongs to a family of proteins comprised of its dimerization partner ARNT (HIF-1 Beta) and the Drosophila proteins PER and SIM. All of these proteins share an N-terminal sequence of ~200 amino acids termed the PAS domain. This receptor, found in a variety of tissues, binds to a specific DNA enhancer sequence and initiates transcription of the mRNA for the cytochrome P-450 (CYPIA1) gene.

  • Q: We have a customer who will use Cat.No. NB100-2289B for Western Blot of mouse samples. The customer will furthermore try to use the antibody for flow cytometry with mouse samples. Therefore, the customer would like to know if you can estimate if the binding site of the antibody at the c-terminus will be inside the protein when the protein is folded in its natural state or on the outside? Do you have any information if this antibody might be suitable for the use in Flow Cytometry? And do you have any information, maybe from blasting the immunogen, if this antibody cross-reacts with any other mouse proteins? First, I thought the part of Localization on your data sheet refers to the protein AHR itself, not only the immunogen? Could you let me know which aa of the c-terminus of AHR are used as an immunogen for this antibody or at least narrow down the region? Could you let me know if this antibody os IgG or IgM and could you let me know which isotype should be used with this antibody?

    A: Localization is First cytoplasmic, upon binding with ligand and interaction with a HSP90, it translocates to the nucleus. This antibody binds to the cytoplasmic side of the protein. This antibody has never been tested for use in flow cytometry, but since it works in ICC, it should work just fine for flow and is not predicted to cross-react with any other mouse proteins. The immunogen used to generate this polyclonal antibody is a very long peptide that falls between amino acids 400-848 of AHR. As a polyclonal, if will recognize epitopes along this entire region. As a rabbit polyclonal, it is an IgG antibody.
