Detection of Cetuximab by Western Blot.
Western blot shows Cetuximab, Rituximab, Brentuximab, and Gemtuzumab biosimilars. PVDF membrane was probed with 0.5 µg/mL of Anti- Cetuximab (Anti-Idiotype) Monoclonal Antibody (Catalog # MAB9626) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Cetuximab at approximately 240 kDa (as indicated). This experiment was conducted under non-reducing conditions and using Immunoblot Buffer Group 1.
Detection of anti-EGFR/ErbB1 (Cetuximab) on A431 Human Cell Line by Flow Cytometry.
A431 human epithelial carcinoma cell line was stained with anti-EGFR/ErbB1 (Cetuximab) (Catalog # MAB9577), then anti-Cetuximab (Anti-Idiotype) (Catalog # MAB9626, filled histogram) or Normal Rabbit IgG Control (Catalog # MAB1050, open histogram), followed by Phycoerythrin-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # F0110).
Specificity of Anti-Cetuximab (Anti-Idiotype) Antibody in Direct ELISA
A panel of biosimilars was coated at 1 μg/mL in 96-well polystyrene plates. Anti-Cetuximab (Anti-Idiotype) Antibody (Catalog # MAB9626, 333 ng/mL) detected Cetuximab. No cross-reactivity with Adalimumab, Rituximab, Trastuzumab, Brentuximab, Gemtuzumab, Ibritumomab, Tositumomab, Solanezumab, Teprotumumab, and Aducanumab was observed.
Detection of Cetuximab by Simple WesternTM.
Simple Western lane view shows Cetuximab and Rituximab biosimilars, loaded at 0.2 mg/mL. A specific band was detected for Cetuximab at approximately 174 kDa (as indicated) using 20 µg/mL of Rabbit Anti-Anti- Cetuximab (Anti-Idiotype) Monoclonal Antibody (Catalog # MAB9626).
