Human Defensin alpha 1 (DEFA1) is 94 amino acids (aa) in length and is cleaved into the following chains: HP 1-56 (aa 39-94), Neutrophil defensin 1 (aa 65-94) and Neutrophil defensin 2 ( aa 66-94). Defensins are secreted peptides that kill microbes by permeabilizing their plasma membrane. DEFA1 has antimicrobial activity against Gram-negative and Gram-positive bacteria. ADP-ribosylation reduces cytotoxic and antibacterial activities, it is also known to enhance IL8 production. Phosphorylation at Tyr-85 is reported in several cancer cell lines and interferes with ADP-ribosylation of DEFA1.

a-Defensin 1 was detected in formalin fixed paraffin-embedded sections of human small intestine using Sheep Anti-Human a-Defensin 1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8198) at 1 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Specific staining was localized to lamina propria in intestinal villi. Recombinant Human a-Defensin 1 protein was serially diluted 2-fold and captured by Mouse Anti-Human a-Defensin 1 Monoclonal Antibody (Catalog # MAB8198) coated on a Clear Polystyrene Microplate (Catalog # DY990). Sheep Anti-Human a-Defensin 1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8198) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).Applications for Human alpha-Defensin 1 Antibody
