Western blot shows lysates of HEK293 human embryonic kidney cell line, SK-Mel-28 human malignant melanoma cell line, and U2OS human osteosarcoma cell line. PVDF membrane was probed with 1 µg/mL of Human CARF Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4195) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for CARF at approximately 61 kDa (as indicated). This experiment was conducted using Immunoblot Buffer Group 1.

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

Background: CARF

CARF (collaborator of ARF) is a component of the ARF-MDM2-p53 pathway. CARF interacts with ARF in the nucleolus accentuating ARF-mediated p53 stabilization and activation. Data also suggests that in the absence of ARF, CARF interacts directly with p53 in the nucleoplasm resulting in a moderate stabilization and activation of p53.
