Detection of Latent TGF¿ beta bp1 by Western Blot

Detection of Latent TGF¿ beta bp1 by Western Blot

LTBP-1 accumulates in CADASIL brain vessels and co-localizes with Notch3-ECD aggregates. a Frozen brain sections from a CADASIL patient and a healthy control containing an arteriole (left panels) or a capillary (right panels) were stained for LTBP-1 and analyzed by fluorescence microscopy. b Frozen brain sections of three CADASIL patients were stained for LTBP-1 and Notch3-ECD and analyzed by confocal fluorescence microscopy. c LTBP-1 co-fractionates with Notch3-ECD. The final ( beta -ME-containing) fractions of sequentially extracted brain vessels from CADASIL patients and controls were immunoblotted for Notch3-ECD, LTBP-1 and beta -tubulin. Shown is a representative blot from two independent experiments. (CAD: CADASIL patient; Ctrl: control). Image collected and cropped by CiteAb from the following open publication (https://pubmed.ncbi.nlm.nih.gov/25190493), licensed under a CC-BY license. Not internally tested by R&D Systems.

Applications for Human Latent TGF-beta bp1 Antibody (35409)

Application
Recommended Usage

Immunoaffinity Purification

This antibody can be used to affinity purify the large latent TGF-beta complex (containing both the 230 and 195 kDa binding protein) and will also pull out “free” binding protein that is not associated with the small latent TGF-beta.

Western Blot

1 µg/mL
Sample: Large latent TGF-beta complex from human platelets
under non-reducing conditions only

Background: Latent TGF-beta bp1

TGF-beta  is synthesized as high molecular weight latent complexes. In platelets, the large latent TGF-beta 1 complex is composed of three components: mature TGF-beta 1 dimer; latency associated peptide; and a latent TGF-beta binding protein (LTBP) (1-3).The cDNAs for four binding proteins (LTBP-1, -2, -3 and -4) have now been cloned (4-5).
