Detection of Mouse Angiopoietin‑like Protein 6/ANGPTL6 by Western Blot.
Western blot shows lysates of Neuro-2A mouse neuroblastoma cell line, 3T3-L1 mouse embryonic fibroblast adipose-like cell line, and J774A.1 mouse reticulum cell sarcoma macrophage cell line. PVDF membrane was probed with 0.5 µg/mL of Rat Anti-Mouse Angiopoietin-like Protein 6/ANGPTL6 Monoclonal Antibody (Catalog # MAB7527) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Angiopoietin-like Protein 6/ANGPTL6 at approximately 52 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Angiopoietin‑like Protein 6/ANGPTL6 in BaF3 Mouse Cell Line.
Angiopoietin-like Protein 6/ANGPTL6 was detected in immersion fixed BaF3 mouse pro-B cell line using Rat Anti-Mouse Angiopoietin-like Protein 6/ANGPTL6 Monoclonal Antibody (Catalog # MAB7527) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rat IgG Secondary Antibody (red; Catalog # NL013) and counterstained with DAPI (blue). Background: Angiopoietin-like Protein 6/ANGPTL6
126-164) and a fibrinogen-like domain (aa 238-456). A conserved Integrin-binding RGD motif is located within the fibrinogen-like domain. This enables ANGPTL6 to promote skin wound healing by mediating the adhesion and migration of keratinocytes, fibroblasts, and endothelial cells. ANGPTL6 also promotes the chemotaxis of vascular endothelial cells resulting in increased vascular permeability and angiogenesis. ANGPTL6 is also secreted by hepatocytes. It inhibits gluconeogenesis in these cells and promotes insulin sensitivity and energy expenditure in mice fed high fat diets. Serum levels of ANGPTL6 are elevated in metabolic syndrome, diabetes, and preeclampsia but are decreased in chronic renal failure. ANGPTL6 is additionally produced by several hematopoietic cell types including megakaryocytes, platelets, mast cells, and uterine NK cells. Mature mouse ANGPTL6 shares 75% and 95% sequence identity with human and rat ANGPTL6.
