ISLR2 (Ig‑superfamily containing LRR protein 2; also SST273) is a 100‑110 kDa member of the LRR family of proteins. It is expressed on developing motor, sensory and interneurons. Mouse ISLR2 is 745 amino acids (aa) in length. It is a type I transmembrane glycoprotein that contains a 571 aa extracellular domain (ECD) and a 132 aa cytoplasmic region. The ECD shows multiple LRRs (aa 21‑231) and one Ig‑like domain (aa 251‑374). Mouse ISLR2 apparently has one alternate start site at Met130. Over aa 20‑590, mouse ISLR2 is 89% and 98% aa identical to human and rat ISLR2, respectively.

Detection of Mouse ISLR‑2 by Western Blot.
Western blot shows lysates of NIH-3T3 mouse embryonic fibroblast cell line and EL-4 mouse lymphoblast cell line. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Mouse ISLR-2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4650) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for ISLR-2 at approximately 80 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.
ISLR‑2 in Embryonic Mouse Brainstem.
ISLR-2 was detected in immersion fixed frozen sections of embryonic mouse brainstem (13 d.p.c.) using Sheep Anti-Mouse ISLR-2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4650) at 1.7 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Specific staining was localized to neuronal processes.
