Detection of His Tag by Western Blot.
Western blot shows lysates of HEK293 human embryonic kidney cell line either non-transfected or transfected with His-tagged human CIQ4 and CHO Chinese hamster ovary cell line transfected with His-tagged BAI-1. PVDF membrane was probed with a 1:4000 dilution of Mouse Anti-His Tag HRP-conjugated Monoclonal Antibody (Catalog # MAB050H). Specific bands were detected for His Tag at approximately 32 and 120 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of His Tag by Simple WesternTM.
Simple Western lane view shows lysates of HEK293 human embryonic kidney cell line transfected with His-tagged human CIQ4 and human MYCOC and CHO Chinese hamster ovary cell line transfected with His-tagged BAI-1, loaded at 0.2 mg/mL. Specific bands were detected for His Tag at approximately 32, 65, and 120 kDa (as indicated) using a 1:1000 dilution of Mouse Anti-His Tag HRP-conjugated Monoclonal Antibody (Catalog # MAB050H). In the blot shown here, the His antibody was used to detect recombinant TcfA, TcfB, and TcfC. The bands are clear with little to no background. Because this is HRP labeled, there is no need for a secondary antibody, which is helpful to speed up the assay. I will use a secondary to increase band intensity.
