Detection of CD4 in Human PBMCs by Flow Cytometry. Human peripheral blood mononuclear cells (PBMCs) were stained with Mouse Anti-Human CD3e APC-conjugated Monoclonal Antibody (Catalog # FAB100A) and either (A) Mouse Anti-Human CD4 Alexa Fluor® 405-conjugated Monoclonal Antibody (Catalog # FAB3791V) or (B) Mouse IgG2AAlexa Fluor 405 Isotype Control (Catalog # IC003V). Preparation and Storage

CD4 is a type I membrane glycoprotein belonging to the immunoglobulin superfamily. It is expressed predominantly on thymocytes and a subset of mature T lymphocytes. CD4 functions in collaboration with the T cell receptor in the recognition of peptide antigens that are presented by class II major histocompatibility complexes. CD4 also has been shown to be a coreceptor of HIV entry and specifically binds gp120, the external envelope glycoprotein of HIV.

This was a 6 color flow cytometry experiment and other antibodies were from another company but only CD4 from rnd systems worked perfectly.
