Detection of Human FoxO1/FKHR by Western Blot. Western blot shows lysates of 293T human embryonic kidney cell line and JEG-3 human epithelial choriocarcinoma cell line. PVDF membrane was probed with 1 µg/mL of Human FoxO1/FKHR Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5939) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for FoxO1/FKHR at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human FoxO1/FKHR by Simple WesternTM. Simple Western lane view shows lysates of 293T human embryonic kidney cell line, loaded at 0.2 mg/mL. A specific band was detected for FoxO1/FKHR at approximately 93 kDa (as indicated) using 10 µg/mL of Sheep Anti-Human FoxO1/FKHR Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5939) followed by 1:50 dilution of HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: FoxO1/FKHR
FoxO1, also called FKHR (forkhead in rhabdomyosarcoma), is a 655 amino acid (aa), 70 kDa, ubiquitously expressed member of the forkhead box O family of winged helix transcription factors. In neurons, it is activated in response to stress, translocating to the nucleus where it promotes apoptosis and blocks proliferation. In insulin-responsive tissues, nutrient abundance triggers phosphorylation by AKT that blocks nuclear translocation and activity. A 60 kDa form, cleaved at R537, has been found in androgen-treated prostate cancer cells. Over aa 353‑655, human FoxO1/FKHR shares 93% aa identity with mouse and rat FoxO1/FKHR.
