Cited-2 is a 270 amino acid protein that functions by binding to the CH1 region of CREB binding protein (CBP)/p300. This binding interferes with certain transcription factors like HIF1 alpha and STAT2 blocking their ability to bind and interact with CBP/p300. Cited-2 knockout mice die due to defects in heart and neural tube development. Mutations in Cited-2 have been reported in patients with congenital heart defects. It appears Cited-2 modulates apoptosis by a p53 independent pathway whereby the details still need to be elucidated.

Detection of Human and Mouse Cited‑2 by Western Blot.
Western blot shows lysates of MCF-7 human breast cancer cell line, HeLa human cervical epithelial carcinoma cell line, and C2C12 mouse myoblast cell line. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse Cited-2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5005) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Cited-2 at approximately 30-35 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Cited‑2 in HeLa Human Cell Line.
Cited-2 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Sheep Anti-Human/Mouse Cited-2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5005) at 1 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red; Catalog # NL010) and counterstained with DAPI (blue). Specific staining was localized to nuclei.
