Western blot shows lysates of K562 human chronic myelogenous leukemia cell line, A431 human epithelial carcinoma cell line, and Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 1 µg/mL of Human/Mouse/Rat CrkL Monoclonal Antibody (Catalog # MAB5127) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for CrkL at approximately 39 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Western blot shows lysates of BaF3 mouse pro-B cell line and Rat-2 rat embryonic fibroblast cell line. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse/Rat CrkL Monoclonal Antibody (Catalog # MAB5127) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for CrkL at approximately 37 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: CrkL

CrkL is a widely-expressed 39 kDa adaptor protein that belongs to the Crk protein family and is abundantly expressed in cells of hematopoietic origin. Through its SH2 and SH3 domains, CrkL initiates interactions with numerous downstream proteins such as CBL, p130Cas, Paxillin, C3G, SOS, PI3K, c-Abl and BCR/Abl. CrkL is involved in such processes as adhesion, migration and the immune response. Furthermore, this adaptor protein plays an essential role in Chronic Myelogenous Leukemia (CML) and is constitutively phosphorylated at Y207 in CML neutrophils by the BCR/Abl kinase.
