HeLa human cervical epithelial carcinoma cell line was stained with Rat Anti-Human/Mouse/Rat GSK‑3 beta Fluorescein‑conjugated Monoclonal Antibody (Catalog # IC2506F, filled histogram) or isotype control antibody (Catalog # IC006F, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: GSK-3 beta
Glycogen Synthase Kinase-3 (GSK-3) is a serine/threonine kinase initially identified as an inhibitor of glycogen synthase. Two isoforms (GSK-3 alpha and GSK-3 beta ) share 85% amino acid sequence identity. GSK-3 beta, inhibited by phosphorylation at S9 by Akt, is involved in energy metabolism, body pattern formation, and neuronal cell development. Depending upon the substrate, phosphorylation may either promote activity (SIK-1; OGT; ZC3HAV1) or downregulate activity (SNAI1; BMAL1; MARK2).
