Q: Why are non-reducing conditions recommended for Human PDGF-B Antibody, Catalog # MAB220, in Western blot?
A: Catalog # MAB220 recognizes a conformational epitope on PDGF-B which can exist as a disulfide-linked homodimer (PDGF-BB) or heterodimer (PDGF-AB). Reducing conditions will break disulfide bonds and destroy the epitope the antibody recognizes. Therefore, non-reducing conditions are needed for Western Blot.
