Detection of Human Serpin B2 by Western Blot.
Western blot shows lysates of U937 human histiocytic lymphoma cell line and HEK001 human epidermal keratinocyte cell line untreated (-) or treated (+) with PMA. PVDF membrane was probed with 1:1000 dilution of Rabbit Anti-Human Serpin B2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8550) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Serpin B2 at approximately 47 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Human Serpin B2 by Simple WesternTM.
Simple Western lane view shows lysates of U937 human histiocytic lymphoma cell line untreated (-) or treated (+) with PMA, loaded at 0.2 mg/mL. A specific band was detected for Serpin B2 at approximately 49 kDa (as indicated) using 1:100 dilution of Rabbit Anti-Human Serpin B2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8550). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.Western Blot Shows Human Serpin B2 Specificity by Using Knockout Cell Line.
Western blot shows lysates of K562 human chronic myelogenous leukemia parental cell line and Seprin B2 knockout K562 cell line (KO) untreated (-) or treated (+) with 300nM PMA for overnight. PVDF membrane was probed with 2 µg/mL of Rabbit Anti-Human Serpin B2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8550) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Serpin B2 at approximately 40 kDa (as indicated) in the parental K562 cell line, but is not detectable in the knockout K562 cell line. HSP60 (Catalog # MAB1800) is shown as a loading control This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
