Detection of Human Tyk2 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line and K562 human chronic myelogenous leukemia cell line. PVDF membrane was probed with 1 µg/mL of Human Tyk2 Monoclonal Antibody (Catalog # MAB3427) followed by HRP‑conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for Tyk2 at approximately 130 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 3.

Tyk2 belongs to the Janus family of intracellular tyrosine kinases that are activated by receptors for many cytokines. Activation of Tyk2 occurs via phosphorylation at two adjacent tyrosine residues, Y1054 and Y1055, within the kinase domain. Tyk2 is required for interferon beta -mediated tyrosine phosphorylation of STAT3 and interferon receptor 1. Mice lacking Tyk2 are susceptible to parasitic infection but resistant to autoimmune arthritis.
