Product Summary
The Quantikine Human VEGF Immunoassay is a 4.5 hour solid phase ELISA designed to measure VEGF165 levels in cell culture supernates, serum, and plasma. It contains Sf 21-expressed recombinant human VEGF165 and antibodies raised against the recombinant protein. Results obtained for naturally occurring human VEGF and recombinant human VEGF121 showed linear curves that were parallel to the standard curves obtained using the Quantikine Human VEGF Immunoassay standards. These results indicate that this kit can be used to determine relative mass values for natural human VEGF.

Preparation and Storage

    Store the unopened product at 2 - 8 °C. Do not use past expiration date.
Background: VEGF
VEGF (Vascular endothelial growth factor) is a potent mediator of both angiogenesis and vasculogenesis in the fetus and adult. It is required for regulating the proliferation, migration, and survival of embryonic endothelial cells and during wound healing and the female reproductive cycle in adults. Pathologically, it is involved in tumor development and tumor vascular leakage. VEGF binds to VEGF R1/Flt-1, VEGF R2/Flk-1/KDR, and Neuropilin-1. Multiple forms of VEGF are generated by alternative splicing and proteolysis.

Assay Procedure
Refer to the product for complete assay procedure.

Bring all reagents and samples to room temperature before use. It is recommended that all samples, standards, and controls be assayed in duplicate.
  1.   Prepare all reagents, standard dilutions, and samples as directed in the product insert.
  2.   Remove excess microplate strips from the plate frame, return them to the foil pouch containing the desiccant pack, and reseal.

  3. Add Assay Diluent
  4.   For Serum & Plasma Samples: Add 100 µL of Assay Diluent to each well.
    For Cell Culture Supernate Samples: Add 50 µL Assay Diluent to each well.

  5. Add Standard, Control, or Sample
  6.   For Serum & Plasma Samples: Add 100 µL of Standard, control, or sample to each well. Cover with a plate sealer, and incubate at room temperature for 2 hours.
    For Cell Culture Supernate Samples: Add 200 µL of Standard, control, or sample to each well. Cover with a plate sealer, and incubate at room temperature for 2 hours.
  7.   Aspirate each well and wash, repeating the process twice for a total of 3 washes.

  8. 200 µL Conjugate
  9.   Add 200 µL of Conjugate to each well. Cover with a new plate sealer, and incubate at room temperature for 2 hours.
  10.   Aspirate and wash 3 times.

  11. 200 µL Substrate Solution
  12.   Add 200 µL Substrate Solution to each well.
  13.   For Serum & Plasma Samples: Incubate at room temperature for 25 minutes. PROTECT FROM LIGHT.
    For Cell Culture Supernate Samples: Incubate at room temperature for 20 minutes. PROTECT FROM LIGHT.

  14. 50 µL Stop Solution
  15. Add 50 µL of Stop Solution to each well. Read at 450 nm within 30 minutes. Set wavelength correction to 540 nm or 570 nm.

  1. Does the Human VEGF Quantikine® ELISA kit (Catalog # DVE00) detect mouse VEGF?

    • No, the kit does not detect mouse VEGF. Specificity testing with recombinant mouse VEGF 120 and VEGF 164 showed no cross-reactivity. For the detection of mouse VEGF, please refer to the Mouse VEGF Quantikine® ELISA kit (Catalog # MMV00).

  2. Does the Human VEGF Quantikine® ELISA kit (Catalog # DVE00) detect porcine samples?

    • Our lab has not tested porcine samples in the kit, however many customers have published use of the kit with porcine samples.  Links to these references can be found in the citations tab by selecting for Porcine in the Species filter.

  3. Does the Human VEGF Quantikine® ELISA kit (Catalog # DVE00) detect human VEGF121?

    • Yes, the kit detects VEGF121.  In specificity testing, 100% cross-reactivity was observed with recombinant human VEGF121

  4. Is the heterodimer VEGF165/PlGF detected by DVE00

    • VEGF165/PlGF is not detected by this kit. Recombinant human VEGF165/PlGF was prepared at 50 ng/mL and assayed for cross-reactivity, with negative results.

  5. Is there a protocol for using cell lysates with the Human VEGF Quantikine® ELISA Kit (Catalog # DVE00)?

    • R&D Systems does not have a validated cell lysate protocol available for this kit.  However, many customers have published use of the kit with cell lysates.  Links to these references can be found in the citations tab.  Generally, the use of detergents in the lysis buffer should be avoided when preparing cell samples.  A better option is to lyse the cells via freeze/thaw cycles or by sonication. 

  6. How long can samples (cell cultures, serum, and plasma) be stored and still be stable for measuring VEGF using the DVE00 Human VEGF Quantikine® ELISA Kit?

Other Experimental Details This VEGF ELISA performs very well and very reproducible. We have used different lot#s, all perform very equally. Well calibrated Standard Protein si provided, curve is almost identical to reference Standard (NIBSC). We usually extend the Standard curve by an additional low calibrator Point (additional 1:2 Dilution of S7), because the Assay still performes well at the low Assay range. Human EDTA-Plasma samples are usually quantifiable and above the detection limit. In general human blood samples matrix should be selected carefully to avoid effects from VEGF release during coagulation.
