Western blot shows lysates of HEK293 human embryonic kidney cell line transfected with N-terminal V5-tagged CBL, N-terminal V5-tagged FSNC4, C-terminal V5-tagged LAMP2B, and C-terminal V5-tagged GALNT2. PVDF membrane was probed with 0.1 µg/mL of Rabbit Anti-V5 Epitope Tag Monoclonal Antibody (Catalog # MAB8926) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). Specific bands were detected for V5 Epitope-tagged at approximately 110 kDa, 34 kDa, 75 kDa, and 65 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Simple Western lane view shows lysates of HEK293 human embryonic kidney cell line transfected with N-terminal V5-tagged CBL and C-terminal V5-tagged GALNT2, loaded at 0.2 mg/mL. Specific bands were detected for V5 Epitope-tagged proteins at approximately 119 kDa and 65 kDa (as indicated) using 1 µg/mL of Rabbit Anti-V5 Epitope Tag Monoclonal Antibody (Catalog # MAB8926). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: V5 Epitope Tag

The V5 epitope tag is derived from a small epitope (Pk) present on the P and V proteins of the paramyxovirus of simian virus 5 (SV5). The V5 tag is usually used with all 14 amino acids (GKPIPNPLLGLDST), although it has also been used with a shorter 9 amino acid sequence (IPNPLLGLD).
