HGK (Hepatocyte kinase-like/Germinal center like-kinase Kinase; also MAP4K4) is a 140 kDa member of the MSN subfamily, MAP4K family of molecules. It is ubiquitously expressed, and intimately associated with TNF alpha signaling. HGK is activated by TNF‑induced c-Jun activation, and blocks both PPAR gamma expression and glucose-stimulated insulin secretion. Human HGK is 1239 amino acids (aa) in length (Accession # O95819) and contains one protein kinase domain (aa 25‑289) and a CHN region (aa 926‑1213). There are multiple splice variants. The antibody product offered by R&D Systems uses a 130 kDa, 1166 aa isoform as immunogen. It shows a deletion of aa 495‑525 and 570‑623, plus an eight aa insertion after His1112, and a GEV insertion after Ala740.

Detection of Human HGK by Western Blot.
Western blot shows lysates of K562 human chronic myelogenous leukemia cell line, HeLa human cervical epithelial carcinoma cell line, MOLT-4 human acute lymphoblastic leukemia cell line, and PC-3 human prostate cancer cell line. PVDF Membrane was probed with 1 µg/mL of Goat Anti-Human HGK Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6027) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for HGK at approximately 140 kDa (as indicated).
