Western blot shows lysates of K562 human chronic myelogenous leukemia cell line, HeLa human cervical epithelial carcinoma cell line, MOLT-4 human acute lymphoblastic leukemia cell line, and PC-3 human prostate cancer cell line. PVDF Membrane was probed with 1 µg/mL of Goat Anti-Human HGK Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6027) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for HGK at approximately 140 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: HGK

HGK (Hepatocyte kinase-like/Germinal center like-kinase Kinase; also MAP4K4) is a 140 kDa member of the MSN subfamily, MAP4K family of molecules. It is ubiquitously expressed, and intimately associated with TNF alpha signaling. HGK is activated by TNF‑induced c-Jun activation, and blocks both PPAR gamma expression and glucose-stimulated insulin secretion. Human HGK is 1239 amino acids (aa) in length (Accession # O95819) and contains one protein kinase domain (aa 25‑289) and a CHN region (aa 926‑1213). There are multiple splice variants. The antibody product offered by R&D Systems uses a 130 kDa, 1166 aa isoform as immunogen. It shows a deletion of aa 495‑525 and 570‑623, plus an eight aa insertion after His1112, and a GEV insertion after Ala740.
