CrkL is a widely-expressed 39 kD adaptor protein that belongs to the Crk protein family and is abundantly expressed in cells of hematopoetic origin. Through its SH2 and SH3 domains CrkL initiates interactions with numerous downstream proteins such as CBL, Cas, Paxillin, C3G, SOS, PI3K, c-Abl and BCR/Abl. It is involved in processes such as adhesion, migration and immune responses. Furthermore, CrkL plays an essential role in Chronic Myelogenous Leukemia and is constitutively phosphorylated by the Bcr-Abl kinase in this disease.

Western blot shows lysates of Jurkat human acute T cell leukemia cell line, BaF3 mouse pro-B cell line, and Rat-2 rat embryonic fibroblast cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse/Rat CrkL Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5127) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for CrkL at approximately 37 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of Human CrkL by Simple WesternTM.
Simple Western lane view shows lysates of K562 human chronic myelogenous leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for CrkL at approximately 44 kDa (as indicated) using 1 µg/mL of Goat Anti-Human/Mouse/Rat CrkL Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5127) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109).
