Western blot shows lysates of Raji human Burkitt's lymphoma cell line. PVDF membrane was probed with 1 µg/mL Mouse Anti-Human PP1 Inhibitor-2 Monoclonal Antibody (Catalog # MAB4719) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). For additional reference, recombinant human PP1 Inhibitor-2 (5 ng) was included. A specific band for PP1 Inhibitor-2 was detected at approximately 34 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: PP1 Inhibitor-2

Protein Phosphatase 1 Inhibitor-2 (PP1 Inhibitor-2), also called PP1 regulatory subunit 2 (PP1R2), I2, and IPP2, binds to PP1 through an RVxF consensus sequence. The complex inactivates PP1, preventing it from dephosphorylating substrates such as glycogen phosphorylase. Unlike PP1 Inhibitor-1, the binding of PP1 Inhibitor-2 is Protein Kinase A phosphorylation-independent. Dissociation of the PP1 Inhibitor-2 complex is enhanced by phosphorylation at T72 by GSK-3, ERK, or CDK. Overexpression of PP1 Inhibitor-2 in mouse heart improves cardiac function without changing organ size.
