MAPK/ERK kinase kinase 3 (MEKK3), also known as MEK kinase 3 and MAP3K3, is a widely expressed 71 kDa Ser/Thr kinase in the SEK, MEK, and MyD88‑IRAK‑TRAF6 signal transduction pathways. It contains a PB1 domain (aa 45‑123), a region with several phosphorylation sites, and a protein kinase domain (aa 361‑621). MEKK3 is involved in tissue morphogenesis, inflammation, and the response to cellular stress. Its activation leads to the activation of NF kappa B following stimulation of IL-1 R, TLR4, TLR8, TNF RI, and the AT1 angiotensin receptor.

Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, Daudi human Burkitt's lymphoma cell line, and A431 human epithelial carcinoma cell line. PVDF Membrane was probed with 1 µg/mL of Mouse Anti-Human MEKK3 Monoclonal Antibody (Catalog # MAB6095) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for MEKK3 at approximately 71 kDa (as indicated). MEKK3 is essential for embryonic vascular development and neonatal brain vascular integrity.(i) Immunoblots show expression of VEGFR2, MEKK3 and T18/S19 phosphorylated MLC2 (MLC2pT18/S19) in freshly isolated BMEC from three Mekk3NCL and three Mekk3 iEC−/− littermate pups as indicated.
