Detection of Human Niemann-Pick Type C2/NPC2 by Western Blot.
Western blot shows lysates of NTera-2 human testicular embryonic carcinoma cell line, human kidney (cortex) tissue, HepG2 human hepatocellular carcinoma cell line, and human skeletal muscle tissue. PVDF membrane was probed with 1:1000 dilution of Rabbit Anti-Human Niemann-Pick Type C2/NPC2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8644) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Niemann-Pick Type C2/NPC2 at approximately 18 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Niemann-Pick Type C2/NPC2 in SK-Hep-1 Human Cell Line.
Niemann-Pick Type C2/NPC2 was detected in immersion fixed SK-Hep-1 human liver adenocarcinoma cell line using Rabbit Anti-Human Niemann-Pick Type C2/NPC2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8644) at 1:150 dilution for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rabbit IgG Secondary Antibody (red; Catalog # NL004) and counterstained with DAPI (blue). Niemann-Pick Type C2/NPC2 was detected in immersion fixed paraffin-embedded sections of human kidney using Rabbit Anti-Human Niemann-Pick Type C2/NPC2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8644) at 1:50 dilution overnight at 4 °C. Tissue was stained using the Anti-Rabbit HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS005) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in tubular epithelial cells.
