The BRCA1 (BReast CAncer gene 1) tumor suppressor protein has many reported functions. In addition to mediating signal transduction in DNA damage and repair responses, BRCA1 regulates transcriptional activity and assists in the preservation of chromosomal stability. BRCA1 is phosphorylated at S1423 by the ATM (Ataxia‑Telangiectasia Mutated) and ATR (ATM-and Rad3-related) protein kinases in response to genotoxic stress.

Detection of Human Phospho-BRCA1 (S1423) by Western Blot.
Western blot shows lysates of U2OS human osteosarcoma cell line mock treated or exposed to 10 Gy IR for 1 hour. PVDF membrane was probed with 0.5 µg/mL Rabbit Anti-Human Phospho-BRCA1 (S1423) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1386) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band for Phospho-BRCA1 (S1423) was detected at approximately 220 kDa (as indicated). The phospho-specificity of this antibody was supported by decreased labeling following treatment with 600 U lambda-phosphatase (lambda-PPase) for 1 hour.
