Polo-like kinase-1 (PLK1) is a serine/threonine kinase that localizes to the mitotic spindle, and is involved in G2 and M phase cell cycle checkpoints. The expression of PLK1 is elevated in many different cancer types. Depletion of PLK1 in mammalian cells results in an inhibition of cell proliferation, decreased viability, and arrest in G2/M phase. PLK1 contains a polo-box domain that binds to phospho-serine and phospho-threonine residues.

Detection of Human PLK1 by Western Blot.
Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line and Raji human Burkitt's lymphoma cell line. PVDF membrane was probed with 1 µg/mL of Human PLK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3804) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for PLK1 at approximately 70 kDa (as indicated).
