|Detection of Rat Phospho-TrkA (Y785) by Western Blot. Western blot shows lysates of PC‑12 rat adrenal pheochromocytoma cell line untreated (-) or treated (+) with the indicated concentrations of Recombinant Rat beta ‑NGF (Catalog # 556-NG) for 5 minutes. PVDF membrane was probed with 1 µg/mL of Human/Rat Phospho-TrkA (Y785) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5479), followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Phospho-TrkA (Y785) at approximately 145 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.|
The receptor tyrosine kinase TrkA binds nerve growth factor (NGF) with low affinity, initiating a signaling cascade that mediates neuronal survival and differentiation. Higher affinity binding of NGF requires a complex formed between TrkA and the p75 NGF receptor. Ligand binding induces receptor dimerization and autophosphorylation on multiple tyrosine residues, including Y490, Y751 and Y785. Phosphorylation of TrkA at Y785 creates a binding site for phospholipase C-gamma (PLC-gamma ), which catalyzes the cleavage of substrate PIP2 to generate the second messengers DAG and IP3.
