Survivin in Human Breast Cancer Tissue.
Survivin was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Goat Anti-Human Survivin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6471) at 1 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC004). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm. Detection of Human Survivin by Western Blot.
Western blot shows lysates of MCF-7 human breast cancer cell line and Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human Survivin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6471) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for Survivin at approximately 18 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Human Survivin by Simple WesternTM.
Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for Survivin at approximately 24 kDa (as indicated) using 10 µg/mL of Goat Anti-Human Survivin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6471) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

The ELISA worked very well. Healthy human serum and plasma samples were not detectable although some disease state samples are detectable.
