Detection of Human Tissue alpha ‑L‑Fucosidase/FUCA1 by Western Blot. Western blot shows lysates of human thyroid tissue and human kidney tissue. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human Tissue alpha ‑L‑Fucosidase/FUCA1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7039) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Tissue alpha ‑L‑Fucosidase/FUCA1 at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Tissue alpha ‑L‑Fucosidase/FUCA1 in HeLa Human Cell Line. Tissue alpha ‑L‑Fucosidase/FUCA1 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Sheep Anti-Human Tissue alpha ‑L‑Fucosidase/FUCA1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7039) at 15 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red; Catalog # NL010) and counterstained with DAPI (blue). Specific staining was localized to lysosomes. Tissue alpha ‑L‑Fucosidase/FUCA1 in Human Liver. Tissue alpha ‑L‑Fucosidase/FUCA1 was detected in immersion fixed paraffin-embedded sections of human liver using Sheep Anti-Human Tissue alpha ‑L‑Fucosidase/FUCA1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7039) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Sheep IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC006). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to lysosomes in hepatocytes. Background: Tissue alpha-L-Fucosidase/FUCA1
Fucosylated glycoconjugates play numerous roles in biological processes, such as development, apoptosis and the immune responses, and are involved in the pathology of inflammation, cancer and cystic fibrosis (1, 2, 3, 4). Tissue alpha ‑L‑Fucosidase, also known as FUCA1, is a lysosomal enzyme responsible for hydrolyzing alpha ‑L‑fucose moieties from glycolipids and oligosaccharides (5). The mature enzyme in the liver has a molecular weight of 200 kDa and exists in the native state as a tetramer (6). Defects in FUCA1 are the cause of fucosidosis (7, 8, 9), an autosomal recessive lysosomal storage disease characterized by the accumulation of fucose-containing glycolipids and glycoproteins in various tissues. Serum alpha ‑L‑fucosidase has been identified as a useful marker for oral cancer (10).
