Overview

The cytoskeleton provides the structural framework and means of movement for the cell. Microtubules control the beating of cilia and flagella as well as chromatin movement during cell division. Actin filaments control vesicle movement throughout the cell and control of the plasma membrane for cell motility and adhesion. Intermediate filaments provide largely static mechanical support for the cell.

beta -I Tubulin in BG01V Human Embryonic Stem Cells. beta -I Tubulin was detected in immersion fixed BG01V human embryonic stem cells differentiated into cardiomyocytes using Mouse Anti-Human beta -I Tubulin Monoclonal Antibody (Catalog # MAB8527) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Desmin in Human Heart. Desmin was detected in immersion fixed paraffin-embedded sections of human heart using 15 µg/mL Human Desmin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3844) overnight at 4°C. Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Vimentin in HeLa Human Cell Line. Vimentin was detected in formaldehyde fixed HeLa human cervical epithelial carcinoma cell line using Rat Anti-Human/Mouse/Rat Vimentin Biotinylated Monoclonal Antibody (Catalog # BAM2105) at 8 µg/mL overnight at 4°C. Cells were stained using the NorthernLights 557-conjugated Streptavidin (orange; Catalog # NL999) and counterstained with DAPI (blue). Specific staining was localized to intermediate filaments. Visualization of F-Actin in HeLa Cells with Phalloidin-TFAX 488. Phalloidin-TFAX 488 (150 nM; Tocris, Catalog # 6622) was used to visualize F-Actin (green) in HeLa human cervical epithelial carcinoma cells. The cells were counterstained with DAPI (Tocris, Catalog # 5748; blue).

This probe only fluoresces upon binding to the microtubules. No wash is required. A no-wash imaging protocol is available.
