G protein-coupled receptors (GPCRs) are the largest family of membrane receptors. As such, drug discovery has focused on this receptor family. Approximately 35% of all FDA-approved drugs target GPCRs. However, this represents only a small proportion of the 800 human GPCRs. This is due, in part, to orphan GPCRs still being understudied, as well as the heterogeneous nature of GPCRs, their endogenous ligands, and their binding sites. Efforts to elucidate the structure, biology, and therapeutic potential of non-targeted GPCRs is ongoing and continues to drive the development of GPCR-based therapeutics.
