Detection of IL-9R by Western Blot Ucma/GRP−/− mVSMCs show increased expression of osteo/chondrogenic markers. Cells were treated with control (CM) or osteogenic medium (OM, with 2.6 mM phosphate) for 12 days. Panels (A–D) qPCR analysis of osteo/chondrogenic gene expression, n = 4–7. Statistical significance was assessed using t-test (Osteocalcin) or ANOVA (E) Western blotting analysis of osteo/chondrogenic markers. Both WT and KO cells were treated with OM. Background: IL-9R
The specific high affinity functional IL-9 receptor complex consists of a ligand-specific IL-9 R( alpha ) type I transmembrane subunit and the common gamma chain that is shared with the receptors for IL-2, IL-4, IL-7 and IL-15. Both the IL-9 R subunit and the common gamma chain are members of the hematopoietin receptor superfamily. Human and mouse IL-9 receptors share 53% amino acid sequence identity, and contain 468 and 533 amino acids, respectively. As a result of alternative splicing, cDNA clones encoding isoforms of IL-9 R, including a putative soluble form, have also been identified. Cells known to express IL-9 R include T cells, neutrophils, mast cells and macrophages.

We used this antibody in an in-house ELISA along with mAb (2901) and protein (290-RNS) to detect IL9R in human serum and plasma. This combination could not detect any IL9R in our samples but generated a good standard curve.
