LINGO‑1 in Mouse Spinal Cord. LINGO‑1 was detected in perfusion fixed frozen sections of mouse spinal cord using Goat Anti-Human LINGO‑1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3086) at 1.7 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Specific staining was localized to the dorsal horn. LINGO-1 (LRRN6A) is a 614 aa transmembrane protein of the leucine-rich repeat (LRR) family, ribonuclease inhibitor subfamily. The four known LINGO proteins contain LRR and IgCAM domains in the extracellular portion and share 44‑61% aa identity. LINGO-1 is restricted to the nervous system and is concentrated in the brain as a component of the NgR1/p75 and NgR1/Taj (TROY) signaling complexes. LINGO-1 negatively regulates neurite outgrowth and myelination. LINGO-1 is highly conserved, showing 99% aa identity between human, mouse and rat.
