Detection of Human/Mouse/Rat Contactin‑3 by Western Blot. Western blot shows lysates of mouse, human, and rat brain tissue. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human/Mouse/Rat Contactin-3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5539) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for Contactin-3 at approximately 150 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

Contactin‑3 in bEnd.3 Mouse Cell Line. Contactin-3 was detected in immersion fixed bEnd.3 mouse endothelioma cell line using Goat Anti-Human/Mouse/Rat Contactin-3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5539) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the Northern-Lights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Background: Contactin-3
Contactin-3 (also BIG-1 in rat and PANG in mouse) is a 150 kDa member of the TAG/F3 subgroup, contactin family, Ig superfamily hierarchy. It is a GPI linked glycoprotein that is expressed on cerebellar Purkinje cells, amygdaloid and thalamic neurons and olfactory granule cells. Its exact function is unclear, but it likely mediates cell-cell interaction and may promote neurite outgrowth. Human Contactin-3 preproprecursor is 1028 amino acids (aa) in length. It contains a 19 aa signal sequence, a 983 aa mature region and a 26 aa cleavable C-terminal prosegment. The mature region shows six C2- type IG like domains (aa 26-593), followed by four fibronectin type III modules (aa 598-992). There is one potential alternate start site at Met128. Over aa 26-992, human Contactin-3 shares 92% aa identity with mouse Contactin‑3.
