|Detection of Human/Mouse/Rat PLC-beta 4 by Western Blot. Western blot shows lysates of DU145 human prostate carcinoma cell line, H4‑II‑E‑C3 rat hepatoma cell line, and Neuro‑2A mouse neuroblastoma cell line. PVDF membrane was probed with 1 µg/mL of Human/Mouse/Rat PLC-beta 4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5128) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for PLC-beta 4 at approximately 140 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.|
PLC-beta 4 (phospholipase C-beta 4; also PLCretA) is a 130 kDa member of the beta -type PLC isozyme family. It is notably expressed in cerebellum and retina. PLC-beta 4 is associated with rhodopsin family GPCRs, where activation of Gq alpha subunits initiates PLC-beta 4 hydrolysis of PIP2. Human PLC-beta 4 is 1175 amino acids (aa) in length. It contains a catalytic X-box (aa 313‑463) and Y-box (aa 565‑681), followed by a calcium-binding C2 domain (aa 688‑786). There are multiple isoforms. One shows an alternate start site at Met114, a second contains a 40 aa substitution for the C-terminal 22 aa, a third shows a 14 aa substitution for the N-terminal 167 aa, and other forms demonstrate combinations of these alternative splice forms. Over aa 75‑250, human PLC-beta 4 is 97% aa identical to mouse PLC-beta 4.
