Western blot shows lysate of human pregnancy sera. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human Pregnancy Zone Protein/PZP Monoclonal Antibody (Catalog # MAB8280) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Pregnancy Zone Protein/PZP at approximately 180 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Recombinant Human Pregnancy Zone Protein/PZP protein was serially diluted 2-fold and captured by Mouse Anti-Human Pregnancy Zone Protein/PZP Monoclonal Antibody (Catalog # MAB82801) coated on a Clear Polystyrene Microplate (Catalog # DY990). Mouse Anti-Human Pregnancy Zone Protein/PZP Monoclonal Antibody (Catalog # MAB8280) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).

Background: Pregnancy Zone Protein/PZP

Pregnancy Zone Protein (PZP), also known as C3 and PZP-like alpha-2-macroglobulin domain-containing protein 6, is a 360 kDa secreted homodimeric glycoprotein, PZP monomer contains a region of decisive functional importance i.e. the bait region. This region serves as a substrate to a variety of proteinases, and cleavage in this domain causes a conformational change of the molecule. Human PZP is a member of the protease inhibitor I39 (alpha-2-macroglobulin) family of proteins.  Highest expression of human PZP is reported in liver, medium expression in ovary, heart and stomach. Also, this protein is found in plasma.  Over amino acids (aa) 26-1482, human PZP shares 56% aa sequence identity with mouse PZP.
