Detection of Human Resistin by Block/Neutralize

Subcellular localization of HBP and resistin in neutrophils.Neutrophils from healthy donors were isolated and stimulated with LPS (50 ng/ml), fMLP (5 μg/ml) or streptococcal M1-protein (1 μg/ml) for 2 h. The neutrophils were immunoflourescently stained for HBP (white), resistin (green) and MPO (red), the cell nuclei were visualized using DAPI (blue). Using scanning laser confocal microscopy, Z-stacks of the cells were acquired and vesicles positive for HBP, resistin or MPO identified using the Imaris image analysis software (processed 3D-Image). The degree of colocalization between the above-mentioned markers in the processed images was then quantified using Imaris analysis software. (A) Shows images of cells from a high responder. Background: Resistin

Human Resistin is an 11-12 kDa member of a small family of inflammation-related cysteine-rich polypeptides. It is named for its antagonistic activity towards insulin (resistance to insulin) and contains five intrachain disulfide bonds that generate a mixture of alpha -helices and beta -sheets. Secreted resistin forms disulfide-linked homodimers that likely oligomerize. Resistin may also form nondisulfide-linked heterodimers with RELM beta. One alternate splice form exists that shows an in-frame 26 amino acid (aa) deletion between amino acids 40 - 65. The significance is unclear. Mature human Resistin is 50% aa identical to human RELM beta and 56% aa identical to mouse Resistin.

    MAB13591 was used as the capture antibody in a sandwich ELISA along with AF1359 as the detection. We also tested BAF1359 as the detection since in comes biotinylated. The standard for the ELISA was 1359-RN. The assay worked excellently! It was sensitive and specific and detected all human samples we tested at a 1:4 dilution.
