Detection of Human AMSH/STAMBP by Western Blot. Western blot shows lysates of HT-29 human colon adenocarcinoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human AMSH/STAMBP Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5650) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for AMSH/STAMBP at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

AMSH/STAMBP in Human Colon. AMSH/STAMBP was detected in immersion fixed paraffin-embedded sections of human colon using Goat Anti-Human AMSH/STAMBP Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5650) at 3 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Background: AMSH/STAMBP
AMSH (Associated molecule with SH3 domain of STAM) is a 50 kDa member of the peptidase M67C class of enzymes. It is widely expressed and serves two functions. First, it promotes receptor recycling by counteracting the effects of ubiquitinating enzymes, and second, it participates in cytokine signal transduction by forming a complex with Jak2/3, STAM and the common beta - and gamma -chains. Human AMSH is 424 amino acids (aa) in length. It contains an NLS (aa 113‑127), two SH3‑binding motifs (aa 195‑198 and 227‑231), and one MPN domain (aa 252‑361). There are five phosphorylation sites at Ser2, 48, 243, 245 and 247. AMSH has one potential splice variant that shows a 17 aa substitution for aa 1‑68. Full‑length human AMSH is 83% aa identical to mouse AMSH.
