BRK (breast tumor kinase; also known as PTK6) is a 50 kDa, intracellular non-receptor tyrosine kinase. It was first identified in human breast tumor cell lines, is frequently over‑expressed in primary breast tumors, and is absent from normal breast tissue. Other sites of expression include colon, small intestine and prostate. Human BRK is 451 amino acids (aa) in length. It contains one SH3 domain (aa 12‑69), one SH2 domain (aa 77‑170) and a Tyr-kinase catalytic domain (aa 183‑444). There are two potential isoforms. The first has a 66 aa extension on the N-terminus, while the second shows the same extension plus a four aa insert after Arg 277. Over aa 1‑70, human BRK is 79% aa identical to mouse Brk.

Brk in Human Lymphoma.
Brk was detected in immersion fixed paraffin-embedded sections of human lymphoma using 15 µg/mL Goat Anti-Human Brk Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3966) overnight at 4 °C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue).
