GLP-2R is a member of the class II glucagon-secretin G protein-coupled receptor superfamily. It is expressed on enteroendocrine cells in the gut where it mediates the positive effects of GLP2 on villus height, crypt depth and intestinal energy absorbtion. After stimulating a cAMP response, lipid raft-dependent internalization of GLP‑2R results in rapid desensitization to GLP2. Extracellular portions of human GLP-2R show approximately 80% amino acid identity with corresponding regions of mouse GLP-2R.
