Detection of Human Latrophilin 2/LPHN2 by Western Blot. Western blot shows lysates of HEK293T human embryonic kidney cell line either mock transfected or transfected with human Latrophilin 2/LPHN2. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Latrophilin 2/LPHN2 Monoclonal Antibody (Catalog # MAB105881) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (HAF018). A specific band was detected for Latrophilin 2/LPHN2 at approximately 110 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.

Detection of LPHN2 in HEK293 Human Cell Line transfected with Human LPHN2 by Flow Cytometry. HEK293 Human Cell Line transfected with Human LPHN2 was stained with Mouse Anti-Human LPHN2 Monoclonal Antibody (Catalog # MAB105881, filled histogram) or isotype control antibody (MAB10041, open histogram), followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (F0101B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (FC005). Staining was performed using our Staining Intracellular Molecules protocol.

Background: Latrophilin 2/LPHN2
Latrophilin 2 (LPHN2) is a G-protein coupled receptor which functions in both cell adhesion and signal transduction. Latrophilin represents a phylogenetically conserved family of receptors which has an unusual structure consisting of two fragments that are produced by proteolytic cleavage of the parental molecule and behave independently in the plasma membrane. LPHN2 is a ubiquitous receptor for alpha-latroxin which is a protein toxin in black widow spider venom that is toxic to vertebrates.
