Detection of MICL/CLEC12A in Human Peripheral Blood Granulocytes by Flow Cytometry.
Human peripheral blood granulocytes were stained with Rabbit Anti-Human MICL/CLEC12A Monoclonal Antibody (Catalog # MAB29461, filled histogram) or isotype control antibody (Catalog # MAB1050, open histogram), followed by Allophycocyanin-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # F0111). MICL/CLEC12A was detected in immersion fixed U937 human histiocytic lymphoma cell line (left panel; positive staining) and Daudi human Burkitt's lymphoma cell line (right panel; negative staining) using Rabbit Anti-Human MICL/CLEC12A Monoclonal Antibody (Catalog # MAB29461) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rabbit IgG Secondary Antibody (red; Catalog # NL004) and counterstained with DAPI (blue). MICL/CLEC12A was detected in immersion fixed paraffin-embedded sections of human spleen using Rabbit Anti-Human MICL/CLEC12A Monoclonal Antibody (Catalog # MAB29461) at 3 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Rabbit IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC003). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in lymphocytes.
