|Detection of Human and Mouse Crk by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, K562 human chronic myelogenous leukemia cell line, NIH‑3T3 mouse embryonic fibroblast cell line, and MOLT‑4 human acute lymphoblastic leukemia cell line. PVDF Membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse Crk Monoclonal Antibody (Catalog # MAB4316) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for Crk at approximately 46 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.|
Crk belongs to a family of adaptor molecules that are widely expressed in numerous tissues. It was originally isolated from a chicken tumor sample, and despite lacking enzymatic activity, this protein is involved in many signal transduction pathways leading to phosphorylation of multiple proteins. Crk forms multi protein complexes via it’s SH2 (aa 13-118) and SH3 (aa 132-192 and aa 237-296) domains leading to activation of downstream molecules involved in growth, differentiation, cell adhesion and migration. Alternate splicing generates an additional isoform that lacks aa 205-304.
