Nephrin (or renal glomerulus-specific cell adhesion receptor; also NPHS1) is a 185 - 200 kDa member of the Ig superfamily. It is expressed on glomerular podocyte membranes, associates with same-cell Neph1, and forms homomeric zipper-like complexes with Nephrin on adjacent podocytes. Mature human Nephrin is a type I transmembrane glycoprotein 1219 amino acids (aa) in length. It contains a 1033 aa extracellular region and a 165 aa cytoplasmic tail. One potential, soluble splice form is known where aa’s 1056 - 1095 are deleted, eliminating the transmembrane region. Over aa 23 - 1029, human Nephrin shares 84% and 89% aa identity with mouse and dog Nephrin, respectively.

Detection of Human Nephrin by Western Blot.
Western blot shows lysates of HEK293T human embryonic kidney cell line and human kidney (medulla). PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Nephrin Monoclonal Antibody (Catalog # MAB42692) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (HAF018). A specific band was detected for Nephrin at approximately 134 kDa (as indicated).
