Detection of Human Neurogenin‑3 by Western Blot.

Western blot shows lysates of human heart tissue, human placenta tissue, human prostate tissue, and human liver tissue. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Neurogenin-3 Monoclonal Antibody (Catalog # MAB3444) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Neurogenin-3 at approximately 27 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Neurogenin-3

Neurogenin-3 (NGN3) is a 23 kDa developmentally expressed nuclear protein of the basic helix-loop-helix (bHLH) family, class B subfamily. bHLH proteins are transcription factors that form dimers to bind DNA. Neurogenin-3 expression is required for all endocrine lineage differentiation in the pancreas, including formation of islet beta  cells. Neurogenin-3 is also neurogenic, controlling the number of dendrites developed by hippocampal neurons. Human Neurogenin-3 shows 76% and 77% amino acid identity with mouse and rat Neurogenin-3, respectively.
